In cardiovascular research, my interests focus on the calcium release mechanism from the sarcoplasmic reticulum involving the ryanodine receptor. We are interested in how this protein is modified under diabetic cardiomyopathy and heart failure particularly by the kinase referred to as the striated muscle preferentially expressed gene (SPEG). To study the interactions between the ryanodine receptor and SPEG, our approaches rely on animal models of diabetes and heart failure, whole organ electrophysiology, single cell calcium dynamics, protein-protein interactions, and biophysical properties at the single protein level. We are interested in using adeno associated virus for in vivo protein delivery, echocardiography for in vivo cardiac function, optical mapping for ex vivo assessment of cardiac properties, confocal microscopy for single-cell calcium dynamic measurements and structural evaluation and planar lipid bilayers for protein biophysics studies.
